Abstract:
Zeolitic Imidazolate Framework-8 (ZIF-8), a novel metal-organic framework, is composed of zinc ions and 2-methylimidazole. It possesses advantages including high porosity, large specific surface area, good chemical and thermal stability, adjustable surface properties as well as pH-induced biodegradability. These advantages equip ZIF-8 with multiple functions, making it widely be used for gas adsorption and separation, catalysis, drug delivery and other fields. Recently, biomacromolecules such as proteins, enzymes and nucleic acids, can be encapsulated by ZIF-8 through in-situ synthesis to form composites. Biomacromolecules are embedded into ZIF-8 crystals or adhered to their surface through cross-linking and covalent bonding. ZIF-8 could serve as a carrier to deliver proteins and enzymes to maintain their biological activities, improving their functional performances and stability. Also, ZIF-8 could transmit nucleic acids, assisting them to express in cells for gene therapy. Therefore, this article reviews the recent research progress of ZIF-8 for biomacromolecules delivery on above issues, and prospects its application in related biomedical fields.
