The development of nanomedicine has expanded the research boundary of biomedicine, especially in the field of precision tumor diagnosis and therapy. Among various nanomaterials, layered double hydroxides (LDH), a typical two-dimensional nanomaterial with good biocompatibility and colloidal stability, large loading capacity, easy functional modification, has attracted extensive attention of researchers in the field of tumor theranostics. In recent years, more and more LDH-based nanoplatforms with exquisite structure, excellent performance and low side effects have been reported in this field. This review introduces the structure and properties of LDH nanoparticles, summarizes the construction strategies of LDH-based nanoplatforms, and summarizes the latest progress of LDH-based nanoplatforms in tumor diagnosis and treatment in the past five years. The challenges and future development strategies for LDH-based nanomedicines toward clinical medicine and precision therapy applications are also discussed. It is hoped that this review will arouse researchers’ wide interest in LDH-based nanoplatforms in the field of tumor theranostics.
